p 53 Gene Mutation in B - Cell Chronic Lymphocytic Leukemia Is Associated With Drug Resistance and Is Independent of MDRl / MDR 3 Gene Expression

چکیده
We studied 5 3 patients with B-cell chronic lymphocytic leukemia (B-CLL) and found mutations of the p53 gene in 15%. Patients with p53 gene mutations were found to have an aggressive form of B-CLL disease characterized by advanced Rai stage, rapid lymphocyte doubling time (LDT), and resistance to chemotherapy. While 27 of 29 treated patients (93%) without p53 mutations achieved a partial remission, only one of seven treated patients (1 4%) with p53 mutations achieved a partial remission (P = .00009). Adjusting for prognostic factors (age, sex, race, and Rai stage), patients with p53 gene mutations had a 13-fold greater risk of death than patients without p53 mutations (P = .013). In addition to examining the clinical relevance

Multidrug Resistance (mdr l ) Gene Expression in Adult Acute Leukemias: Correlations With Treatment Outcome and In Vitro Drug Sensitivity
Resistance to multiple chemotherapeutic agents has been related to the production of P-glycoprotein, a trans-membrane drug efflux pump that is encoded by the multidrug resistance (MDR) gene mdrl. To investigate whether mdrl could be involved in clinical resistance to chemotherapy in acute leukemias, we have analyzed retrospectively the RNA from adult acute leukemia cells by slot-blot hybridizat...
متن کاملAnalysis of HLA-G Gene Expression in B-Lymphocytes from Chronic Lymphocytic Leukemia Patients
The human leukocyte antigen G (HLA-G) molecule exhibits limited tissue distribution, low polymorphism and alternative splicings that generate seven HLA-G isoforms. HLA-G exerts multiple immunoregulatory functions. Recent studies indicate an ectopic up-regulation in tumor cells that may favor their escape from anti-tumor immune responses. This study it is an effort to clarify the presence of HLA...
متن کاملEffect of curcumin on growth inhibition and gene expression reduction in chronic lymphocytic leukemia cells (CLL-CII)
Introduction: Chronic lymphocytic leukemia (CLL) is the most frequent leukemia in adults living in Western countries. with a diverse clinical course that due to life expectancy less than two years between patients finding ,new therapeutic strategies is essential for it. Curcumin as the biologic active of turmeric, has many biological properties such as anti-inflammatory, anti-oxidant, anti-diab...
متن کاملAnalysis of Expression Of SIRT1 Gene In Patients With Chronic Myeloid Leukemia Resistant To Imatinib Mesylate
Background: Chronic myeloid leukemia is a clonal myeloproliferative disease which is characterized by bcr/abl translocation. With the emergence of tyrosine kinase inhibitors such as imatinib mesylate, significant improvement has been made in treatment of this disease. However, drug resistance against this medicine is still an obstacle. SIRT1 is a gene with deacetylase activity which has been de...
متن کاملChronic lymphocytic leukemia, biology, new diagnosis and treatment: review article
Chronic lymphocytic leukemia (CLL) is a malignancy of B CD5+cells and is the most common type of leukemia in adults. The disease is more common in men over 50 years in western countries. CLL is associated with defective apoptosis in B cells. CLL was traditionally regarded as a disease that occurs before naïve B cells meet the antigen in the lymph nodes. Laboratory diagnosis requires white blood...
